Intended use

This ELISpot Path kit is intended for the enumeration of cells secreting human IFN-γ in response to BK virus (BKV) peptides. For research use only. Not for use in diagnostic procedures.

Peptide pool composition
PepPool: BKV (CD4 and CD8), human contains 65 synthetic peptides derived from the BKV large T protein, small T protein, and VP1 protein. 50 of the peptides are MHC class I-restricted and 15 are MHC class II-restricted. The peptide pool is designed to stimulate T cells with a broad array of HLA class I supertypes. Sequence similarity to corresponding JC virus (JCV) peptides ranges from 60% to 100%, with a median sequence similarity of 80%.

Human PBMC (250,000 cells/well) were incubated for 20 h in the absence or presence of PepPool: BKV (CD4 + CD8), human, and the number of cells secreting IFN-γ was analyzed by ELISpot. Analysis was done with Mabtech IRIS.

IFN-γ

Analyte description

Interferon-γ (IFN-γ) is the only type II interferon. This proinflammatory cytokine is secreted by activated T cells and NK cells. It activates macrophages and endothelial cells and regulates immune responses by affecting APCs, T cells, and B cells. Production of IFN-γ by helper T cells and cytotoxic T cells is a hallmark of the Th1-type phenotype. Thus, high-level production of IFN-γ is typically associated with effective host defense against intracellular pathogens.
